I've been re-reading the whole thread and I'm afraid I may have made a mistake.

One of gen 1's best friends was the IF of his sister. His sister was best friends with her IF but their relationship deteriorated so that they were acquaintances only. I'm worried that since the IF was best friends with someone in the household I'm not allowed to count him as unique. Or can I count him?

And some questions,
1)Are we allowed to move out the spouse of the current heir after the next gen is born?
2)Are there any restrictions on using the Philosopher's Stone to raise ghosts and then using ambrosia to bring them back to life?
3)Is it okay to sell the cameras that appear in a sim's inventory at the start of the game?

I can't answer your first question, but for the other three:

1. Yes
2. Yes
3. For your founder(s), I think you have to sell them before you set their funds to 4,000, but that you can sell the cameras that come with whoever you move in. Someone should confirm, though.

Does anyone know how to change lifespans without losing/gaining days? I put lifespan on medium for my 4x4 and switched to normal for my practice ID. When I made the switch, the founder had 16 days to age up instead of 21.

No, you can't count him as unique.  Sorry :(
Rule:  These Best Friends must be totally unique to a given heir and cannot be Best Friends with anyone else in the household.

Yes.

No restriction. 

3)Is it okay to sell the cameras that appear in a sim's inventory at the start of the game?

There is no restriction to selling the camera, so you are allowed to do it.  You can sell it after you set funds to 4000.  Yes I know this sounds contradicting to starting poor, but people that choose to start with townies are allowed to sell furnitures too.

So when you made the switch "back" to medium, the founder has 16 days to age up instead of 21?  I never had any problem with losing/gaining days while switching lifespan.

Yes. The rule states the household is allowed 4 trips, so anyone in the household counts against that.

I'm a bit unclear on what is and isn't allowed. What I'm getting from this thread, non-heirs can invite anyone into the household, they can't be spouses but their children born into the household can marry heirs, is this right?

So I'm going to restart mine, since it's picking up glitches all over the place like mad, even with the permitted move to fix the aging glitch (my favorite one is when moving, the save-under-new-name overwrites the old-with-a-different-name).

And as I'm going through the rules again, and I want to confirm that the moving heir's spouse can do the same careers as previous world founders. But future moving spouses cannot do the same careers/skills/challenge completions (even by accident) because, when the move, they then become founders of the new world, and neither founder can do any of those that have been (or will be) completed by an heir.

Moving heir spouses can complete the same careers, skills, etc. as the original founders and other moving spouses just not the same ones as any heirs, because, yes, they become founders. Is that what you were asking? I'm not really sure what I'm saying is clear at all. Sorry.

Nope, you confirmed it for me.  :) I figure I'll just have all the moving heirs' spouses do the same as the original founders. Gen4's spouse somehow completed master chemist in the logic skill, but still had an option to discover a potion, I found out today... >:(

I get that all the time.  I thought it was an odd feature of the in-game town move but maybe it's a bug.  I'm paranoid enough to do several saves under different names just before a move, so it's only the last one that gets overwritten.  Weird, though.

So I looked over the list of acceptable collection stuffs very carefully, and didn't see it, so wanted to check - items that are made with the Prism Studio/glass blowing thing, are those items approved?
