Does the lot have to be flush with the road?

I want to place a lot but it doesn't go flush with the road.  Does it have to?  Can there be a gap of one or two grids sizes from the lot and the road?

Does it matter if it is a home or commercial lot?  I want to put a business in MF, but the grid doesn't go flush against the road.

No, you can place a lot miles away from the road, as long as the area is routable.
